The fire broke out at the house in Lytham Road , Freckleton, at about 23:20 GMT on Saturday.
Reece Smith, 19, four-year-old twins Holly and Ella Smith and two-year-old Jordan Smith were taken to hospital but all afterward passed away.
Their mother, considered to be 36-year-old Michelle Smith, was led away from the sight unharmed.
The four siblings were pulled insensible from the bungalow by firefighters and tragedy services attempted to save them at the sight.
Lancashire Constabulary said police were keeping an open mind as to what might have reasoned the fire.
They are now implementing a joint inquiry with the fire service.
Mrs. Smith's father, Keith Smith, 56, said it had been his daughter's birthday on Saturday.
He said she had a whole of nine children.
"It's very dreadful, these are such sad condition," he said.
"I don't know something about what taken place last night. They have lived here for at least two-and-a-half years - this was their grandmother's previous home."
Mr. Smith added: "I last saw them all just after Christmas.
"I can't imagine of any cause why this has taken place. It has just come as a big upset."
Det Andy Murphy, of Lancashire Constabulary, said officials were still endeavoring to set up what had caused the fire.
He would not remark on whether a party was held at the address on Saturday night, however, he said other people had been in the house at the time.
"We require to evaluate how many people were there and decide what occurred," he said.
Mr. Murphy added: "This is a totally disastrous mishap and our considerations are with the family at this very tough time."
The fire affected bedrooms of the house in the village, which are quite a few miles west of Preston .
From the outside there was no noticeable loss caused by the fire, but the sight has been cordoned off.
Friends and family have begun sending off floral compliments near the house.
Phil Cox, group manager for Lancashire Fire and Rescue Service, said as squads were en way they had got calls saying people were trapped inside the property, including several little children.
"Once the crews received it considered quite a disordered scene with a number of people outside quite upset," he said.
"We sent in six firefighters who saved four people and also led out one other person.
"We had detectives, fire officers and paramedics at the sight working on saving the people brought out before they were taken to hospital.
"We would like to send our sympathies to the family.
"There was a realistic amount of smoke at the time, we were faced with quite a profound smoke-logged building."
Family friend Carolyn Milburn said Reece was the eldest child and Jordan the youngest of Michelle Smith.
She said: "It's knocked us all for six.
"They were all pretty, all of them.
"Lovely toddlers well behaved, well brought-up, always glad.
"Michelle is a good mum. She loves her kids.
"I haven't spoken to her yet but her and her mother Christine are both in a state."
She further said, "She is in bits over this. The twins were lovely, beautiful children. They were all smashing kids."
One of the floral compliments left outside the house read: "To the family and loved ones of Reece, Holly, Ella and Jordan, we are regretful to listen about the painful and tragic loss of four adolescent lives.
"Our considerations and prayers are with you all. Love from a Freckleton family xxx."
